Monit can watch the filesystem flags for change already, thus can check that the filesystem was not remounted without quota support. It could be also possible to implement the test which will check that the quota enforcement work in general. On the other side, if you mean per-user quota provisioning and reporting then i'm not sure whether monit should do this - per-user quota management is probably out of scope.
In general, it is possible to use external script for monitoring various services which are not supported by monit currently. There is nice trick developed by Pavel Urban, see for example: http://lists.nongnu.org/archive/html/monit-general/2007-01/msg00008.html
