I've tried an older version of plex86 (downloaded in June 2002). With
this version, I was able to boot from a DOS floppy disk.
If you want do do so, make sure you have access rights (read+write) to
/dev/fd0 and set up the config file like the following:
bochs-opt floppya: 1_44=/dev/fd0
Maybe you can also boot from real harddisks, I never tried.
Booting from CD is AFAIK not possible.
BTW: When I booted DOS from the Setup disk, plex86 didn't recognize disk
changes, so I had to copy the three disks into the harddisk image and
uncompress the files manually. Is this problem known (or even fixed)
and/or is there a method how plex86 and the guest OS will notice disk
changes?

Linux - life is too short for reboots.
