Yes, you're right: I think the file you are referring to is the 512-byte copy of your Slackware partition boot sector. You need to copy it back to Windows every time you reinstall your boot loader.

b) how I can solve this problem.

Just put your Slackware 10.0 CD in and boot like this:

boot: linux root=/dev/<your slackware partition> ro

I haven't got my Slack at hand, so I am not absolutely sure about the syntax. Anyway, you should find the information you need in the brief text immediately above the boot prompt.
It should start your current system with the default kernel.
Maybe something won't work, but should be enough for you to access your partitions and extract the boot sector as detailed in the above URI.
